Basement Membrane Matrix, Growth Factor Reduced: Enhancing Organoid and 3D Cell Culturing

Date:08-27-2026

Basement membranes are specialized extracellular matrix sheets that serve as interfaces between different types of cells and their surrounding stroma. They play a crucial role in tissue organization, affecting cell adhesion, migration, proliferation, and differentiation. Basement Membrane Matrix, Growth Factor Reduced, offered by Biogradetech under the catalog number A-MGL01, is a soluble form of basement membrane purified from Engelbreth-Holm-Swarm (EHS) tumor. This matrix is composed of laminin, collagen IV, entactin, and heparin sulfate proteoglycan, and it provides a reconstituted basement membrane when incubated at 37°C.

Product Description

Basement Membrane Matrix, Growth Factor Reduced, is derived from mouse tumor sources. It is available in two variations: one containing phenol red, which appears yellow-pink, and another phenol red-free variant, which is translucent light yellow. The standard Basement Membrane Matrix is in a liquid state at 4°C, while the high concentration variant forms a semi-gel when kept at 0°C for an extended period. The protein concentration ranges from 8 to 26 mg/mL, and the endotoxin level is ≤4.5 EU/mL.

Intended Use

In addition to applications where Standard Basement Membrane Matrix can be used, the Growth Factor Reduced variant is particularly suitable for demanding research applications. It is commonly used to optimize signaling in tubular bone cells and to study gene expression in primary mouse mammalian epithelial cells, reducing background signals induced by growth factors.

Usage Instructions

Thick Gel Method:

Thaw Basement Membrane Matrix as instructed above.

Mix the solution by gently pipetting it up and down to avoid introducing air bubbles.

Pipette 200-300 μL per cm2 onto the growth surface.

Incubate the coated object at 37°C for 30 minutes.

The coated objects are now ready for use.

Thin Layer Method (non-gelling):

Thaw Basement Membrane Matrix as instructed above.

Mix the solution by gently pipetting it up and down to avoid introducing air bubbles.

Dilute the Basement Membrane Matrix to the desired concentration in cold serum-free medium. A 1:100 dilution is recommended for the propagation of primary cells.

Add a sufficient amount of the solution to cover the entire growth surface area. A volume of 300 µL per cm2 is recommended.

Incubate the coated object at room temperature for one hour.

Aspirate the coating solution and immediately plate the cells. Ensure that the coated surface does not dry out.

Conclusion

Biogradetech’s Basement Membrane Matrix, Growth Factor Reduced, provides researchers with a valuable tool for organoid and 3D cell culturing. Derived from mouse tumor sources, this matrix offers laminin, collagen IV, entactin, and heparin sulfate proteoglycan components. It can be used in various applications that require different thicknesses and concentrations, enabling researchers to optimize
