Human T-Activator CD3/CD28 Beads: Facilitating Human T Cell Activation and Expansion

Date:08-27-2026

Human T-Activator CD3/CD28 Beads, available from Biogradetech under the catalog number C-BETA2001, are specifically designed for the separation and in vitro expansion of human T cells. These beads find applications in various T cell culture technologies, including CAR-T cell therapy. By leveraging the power of anti-human CD3 and anti-human CD28 antibodies conjugated to magnetic beads, Human T-Activator CD3/CD28 Beads provide a simple and effective method for isolating and expanding human T cells.

Product Information

Catalog: C-BETA2001

Reactivity: Human

Concentration: 2 × 108 beads/mL

Particle size: 4.5 μm

Endotoxin: 1 EU/mL

Usage: In vitro T cell activation and expansion in enriched T cell or PBMCs

Formulation: Phosphate-buffered saline (PBS) containing Human Serum Albumin (HSA), pH 7.4

Stability: 24 months

Materials Required

Human T cell culture media.

Human cytokines for optimal expansion, such as IL-2, IL-7, IL-15.

Protocol

Wash Human T-Activator CD3/CD28 Beads

Resuspend the beads in the tube and vortex or tilt and rotate to ensure proper mixing.

Transfer the desired volume of beads to a new tube.

Add an equal volume of PBS (containing 1% HSA) for resuspension.

Place the tube on a magnet for 1 minute and discard the supernatant.

Remove the tube from the magnet and resuspend the beads in the same volume of PBS (containing 1% HSA).

Separate CD3+ T cells

Gently resuspend Ficoll-isolated PBMCs in PBS (containing 1% HSA) and adjust the cell density.

Add the washed beads to the PBMCs in the appropriate ratio.

Rotate the samples and incubate at room temperature.

Dilute the mixture of beads and cells with T cell culture media or PBS (containing 1% HSA) and place the tube on a magnet.

Remove the supernatant and resuspend the mixture in T cell culture media containing IL-2.

T cell activation and expansion

Count the number of T cells daily starting from day 3.

Dissociate the beads and cells by gently blowing the cell suspension during the later stages of cell expansion.

When CD3+ T cells exceed 1 × 106 cells/mL, dilute the cells with T cell culture media containing IL-2.

At the end of the culture (day 9-14), count the cells and remove the beads using magnets.
